Take a plane. It's much safer, quicker, and you can usually get one for about $100. A bus ticket with a reputable company will run you $50 and it will be a 10 hour drive.

There was a white American girl who was travelling alone who took a bus from Medellin to Bogota this month (May 2012). She got there safely and said it was fine. I don't think it's too dangerous. I'm not sure of the company she used but I know she picked it off of google.

There is a brand new airline in Colombia that flies new Airbus 320's at deep discount prices. Bogota to Medellin Rio Negro airport was US$35 one way. Don't take the bus - it's safe but long. Fog frequently closes the Rio Negro airport in the early morning.
